Location of Heme Synthesis

The first and last steps of heme synthesis occur in the mitochondria. In the mitochondria, succinyl-CoA and glycine combine to form δ-aminolevulinic acid (ALA), which is the initial precursor of heme synthesis. ALA then moves to the cytoplasm where several intermediate steps take place. These steps involve various enzymes and intermediates that ultimately lead to the formation of heme. The final stages of heme synthesis occur back in the mitochondria, where heme is fully synthesized.

Therefore, heme synthesis is a coordinated process that involves both the cytoplasm and mitochondria of the cell, each playing a specific role in the synthesis of this vital molecule.
